چکیده

Background: Ileal bile acid binding proteins display different degrees of binding cooperativity. Results: The structure of a low cooperative protein complexed with bile salts was determined. The protein was mutated to enhance its cooperativity. Conclusion: The cooperative binding requires few latch residues that stabilize an H-bond network. Significance: Knowledge of the determinants of cooperativity in protein carriers is key to understand bile acid trafficking.

Energetics by NMR: site-specific binding in a positively cooperative system.

Proteins with multiple binding sites exhibit a complex behavior that depends on the intrinsic affinities for each site and the energetic communication between the sites. The contributions from intrinsic affinity and cooperativity are difficult to deconvolute using conventional binding experiments that lack information about the occupancies of individual sites. Here, we report the concerted use ...

Cooperativity in biological systems

Living organisms can sense and respond to external and internal stimuli. Response isdemonstrated in many forms including modulation of gene expression profiles, motility, secretion, cell death, etc. Nevertheless, all forms share a basic property: they depend on sensingsmall changes in the concentration of an effector molecule or subtle conformational changes ina protein and invoking the appropri...

UDCA to partially block the transmembrane apical Na

This article is available online at http://www.jlr.org UDCA to partially block the transmembrane apical Na + dependent bile acid transporter ( 7 ). Thus far, no intracellular receptor for UDCA has been reported. The farnesoid X receptor (FXR ) is a nuclear receptor that binds to and is activated by bile acids ( 8–10 ). FXR forms heterodimers with the retinoid X receptor (RXR ) and regulate...

Temperature dependence of backbone dynamics in human ileal bile acid-binding protein: implications for the mechanism of ligand binding.

Human ileal bile acid-binding protein (I-BABP), a member of the family of intracellular lipid binding proteins plays a key role in the cellular trafficking and metabolic regulation of bile salts. The protein has two internal and, according to a recent study, an additional superficial binding site and binds di- and trihydroxy bile salts with positive cooperativity and a high degree of site-selec...

The X-ray structure of zebrafish (Danio rerio) ileal bile acid-binding protein reveals the presence of binding sites on the surface of the protein molecule.

The ileal bile acid-binding proteins (I-BABPs), also called ileal lipid-binding proteins or gastrotropins, belong to the family of the fatty acid-binding proteins and play an important role in the solubilization and transport of bile acids in the enterocyte. This article describes the expression, purification, crystallization, and three-dimensional structure determination of zebrafish (Danio re...
